Herceptin functionalized microfluidic polydimethylsiloxane devices for the capture of human epidermal growth factor receptor 2 positive circulating breast cancer cells

Building on recent breakthroughs in the field of microfluidic-based capture of rare cancer cells circulating in the blood, the present article reports on the use of Herceptin functionalized PDMS devices designed to efficiently capture from blood cancer cells, overexpressing the tyrosine kinase human epidermal growth factor receptor (HER2). The identification of patients overexpressing HER2 is critical as it typically associates with an aggressive disease course in breast cancer and poor prognosis. Importantly, HER2 positive patients have been found to significantly benefit from Herceptin (Trastuzumab), a humanized monoclonal antibody (MAb) against HER2. Disposable PDMS devices prepared using standard soft lithography were functionalized by the plasma polymerization of an epoxy-containing monomer. The epoxy-rich thin film (AGEpp) thus created could be conjugated with Herceptin either directly or through a polyethylene glycol interlayer. The properties and reactivity toward the monoclonal antibody conjugation of these coatings were determined using x-ray photoelectron spectroscopy; direct conjugation provided a good compromise in reactivity and resistance to biologically nonspecific fouling and was selected. Using the breast cancer cell line SK-BR-3 as a model for cells overexpressing HER2, the immunocapture efficacy of the Herceptin functionalized PDMS was demonstrated in model studies. Validation studies confirmed the ability of the device to efficiently capture (～80% capture yield) HER2 positive cells from full blood.  相似文献

Expectations versus reality: the case of Liberal Studies in Hong Kong’s new senior secondary reforms

This article reports the findings of a five-year longitudinal study investigating the introduction of Liberal Studies in the context of Hong Kong’s new senior secondary educational reforms. The aim of this empirical research, which adopted diachronic analysis to allow comparisons over time, was to compare teachers’ and students’ perceptions of the Liberal Studies curriculum before and after its introduction. Methodologically speaking, the study was informed by documentary inquiry, with approximately 560 newspaper articles and government reports collected to identify the issues surrounding the curriculum debate. Questionnaire-based surveys conducted in the pre- and post-reform periods revealed a sharp contrast between expectations and reality for both participant groups following the curriculum’s introduction. Whilst the students demonstrated favourable attitudes towards Liberal Studies before studying the subject, a certain degree of disappointment was expressed after the new curriculum came into effect. In follow-up interviews with teachers, they expressed a lack of confidence in teaching such a multi-disciplinary subject and criticised the overly ambitious nature of the curriculum. Liberal Studies, as currently taught, runs the risk of falling out of favour with Hong Kong secondary school teachers, and the question of whether it will play an increasingly important role in promoting patriotic education after the demise of Moral and National Education is deserving of research attention.  相似文献

From learning to labour to learning for marginality: school segregation and marginalization in Swedish suburbs

In this article, using data from ethnographic research, we try to present some glimpses of the way education is described as an experience and possibility ‘from below’, by pupils who grow up and study in schools in the most segregated and territorially stigmatized suburbs on the outskirts of our major cities. What we feel they describe is an experience of schooling for surviving the social and economic consequences of curtailed citizenship in a post‐industrial society rather than one of schooling that offers possibilities of integration and full citizenship or social transformation. Our findings have significant policy implications in this respect. Sweden has historically pursued projects aimed at educational inclusion but has recently taken a significant turn toward neo‐liberalism and educational consumerism, since which time various disadvantaged groups have become increasingly concentrated compared with others in under‐achieving schools in an economically threatened public sector. The education of children with a medical condition represents a unique educational context. The key educational factors that can help these children continue their education despite the burdens associated with their illness were discussed and analysed by a pool of experts for an EU funded project. In this context, relationships, making sense and constructing knowledge, assuming roles in front of others, metacognition, individualities and inter‐institutional communication emerged as the 6 Key Educational Factors (KEF) that are crucial for the education of this vulnerable population. The implications of the KEFs for home and hospital education are discussed, with a particular focus on practices that meet the relational and communicational needs of these children. Specific recommendations for the practice, policy, and research regarding these KEF within this unique educational context are presented.  相似文献
